We are looking at the molecular mechanisms that establish in the brain after chronic stress and that result in resilient behaviour. These mechanisms include epigenetic imprints, expression of non-coding RNAs and changes in the transcriptional and translational program of the cell. We focus on chronic social defeat stress on one hand and on different kinds of early life stress on the other and are also interested in the elucidation of putative inheritance of resilient behaviour from one generation to the other. We are using mouse models including the Ribotag and the SUN-GFP mouse that allow cell-specific sorting of affected cells and combine these with single cell RNA sequencing, Bi-sulfite sequencing, ATAC sequencing and mass spectrometry. Epigenetic modulators and mTOR kinase inhibitors are used to modulate resilient behaviour. Furthermore, in human, cortical organoids we are looking at changes in X-inactivation in the developing female brain as a potential mechanism underlying gender specific behaviour.
